I am doing a Qiagen giga prep on a plasmid that I recently cloned. I didn't realize how long the column would take to drain via gravity, and I have to catch a train. Is it possible to stop the prep at the isopropanol precipitation stage and not pellet it until Monday? And if so, what temperature should i store the plasmid/isopropanol mixture? note: the Qiagen protocol says that it is possible to leave the prep overnight (but not longer) at the previous step in which the DNA is eluted in Buffer QN.
absolutely. This is a good way to save dna and I am not even sure there is a limit to how long you can store this way. But overnight in the refrigerator is surely fine...You can also leave at room temp to make sure no salt ppts out.
yep should be OK. we left our samples overnight at 4C many times (both with and without isopropanol), and after centrifugation next day the yields were about right. only once we left the samples at 4C for 2-3 days, and the yields seemed to be somewhat below expectation, but just slightly
