I was trying to revive a freeze-dried culture of thermophilic carboxydotrophs, but accidentally the pH of the culture in which I inoculated the freeze-dried culture was much above the optimum for the organism. (optimum=6.8-7.0, revival medium=8.8)

I realised that there was something wrong with the medium when I observed no growth after 16 hrs (Td=2hrs). When I tested the left over medium from the same batch, I found that the pH was 8.8.

I was wondering if it would be possible to salvage the inoculated revival culture by taking the vial out of the incubator, adding a calculated amount of acid to bring down the pH to optimum range, and putting it back.

Is it possible that the culture will revive on doing this? Or would the initial inoculum have completely degraded by now?
