I'm culturing Caco-2 cells in 96 wells plastic microplate and 24 wells Transwell inserts to work on differentiated monolayers. I've heard some information as Caco-2 differentiation is better on Transwell and Caco-2 polarization might be problematic on glass or plastic support as cells have no space on basolateral side, but i haven't found articles which clearly talk about it.
Do you have some articles to share about this question ?
Hi, I've always used inserts with my Caco-2 cells mainly because I was interested in sugar transport across a polarised / differentiated monolayer. The most important bit when growing them 2D is using ECM on your chosen surface. The link below is to a nice paper looking at different ECM types on plastic & glass and their effect on differentiation. In my experience, adding butyrate to the media also speeds up differentiation.

However I've already done my experimentation for a toxicological evaluation
- One experiment on 96 well microplate for dose-effect response on a wide concentration range ( easier and cheaper than Transwell)
- A second one with selected concentrations (choose on the first results) on Transwell.
My observation is a difference between cells cultured in plate and in Transwell about cytotoxicity induce by my product.
I try to understand why and may be it's a difference in transporter expression between the two culture support ? Medium was exactly the same in both experimentation.
