My lab has been brought an array of C-4 I cell line. The cells did not succeed in attaching to the bottom of the flask and remained floating in the media. Although we followed the protocol for this type of cells, which we obtained from the source. Does anyone have any experience with culturing these cells and if so, would you mind sharing that knowledge? Thanks a lot in advance!

In my experience, if the cells adherent and you found them floating, it is more likely the cells are dead. You need to check CO2 percent, media, and the FCS. Also, if the cells are growing (not dead) you can notice changing the colour of the media toward yellow, and if the red colour stay as it means, there is no growth.

I had some cells once that would not attach. I tried different tissue culture flasks and other things but it ended up being that the cells would not attach in one of our incubators. When I grew them in a different incubator they attached just fine. There must have been type of vibration or something that prevented them from getting comfortable. I was surprised by this and never would of suspected. Never stop questioning.
