Is there a general rule to follow as far as number of nucleotides and the sequence is concerned? Or is there a strict rule to follow depending on restriction enzyme to be used?
Buffer sequences for efficient restriction digestion.
There is no general rule as some enzymes cut right up to the recognition site and a few cut up to 10 bases away from it so you should check the New England Biolabs website which specifies this information for many enzymes. In general 3 extra bases is enough for almost all enzymes
