I’ve been performing multiple peripheral blood lymphocyte cultures for metaphase chromosome preparation. The cell density seems fine at the time of harvesting, but the number of metaphase spreads is consistently low.

Additionally, the few metaphases I do get are highly overlapped and not well spread.

Does anyone have insight into what might be causing this issue?

Any suggestions for optimizing metaphase yield and spreading would be greatly appreciated.
