Hello, I'm conducting the biofilm formation experiment using PAO1(-GFP).

The problem is, biofilm is formed only on liquid-air phase, not on the bottom of wells.

My conditions are below:

PAO1 OD 0.03, 0.5ml w/M9 + OD 0.03, 0.5ml w/LB (total 1ml/well)

plate incubation: 120rpm, 20h, 37 Celsius degree

I already tried 0rpm, 20h or 180rpm, 6h but the problem still remains.

How can I do with this?
