Hi everyone, we performed Caco-2 Permeability (Bi-directional) assay for different compounds, and when we measured Papp (B-A) and Papp (A-B), in most of the tested compounds we found only Papp (B-A) while Papp (A-B) was below the LOD. I would like to know if in this case is it possible to calculate efflux ratio? And is it possible with this test to observe only Papp (B-A) and not Papp (A-B)? Many thanks to everyone.
You obviously can't calculate an Efflux Ratio, but you have some conclusions to make. One is that the Ppassive must be very slow assuming your collection intervals are long enough and your bioanalytical sensitivity is sufficient. One example of this behavior is sulfasalazine where it is known to have an active basolateral-in transporter that is yet (to my waning knowledge) unidentified and an active apical-out (P-gp) transporter.
