I'm working with PBMCs from patients and am interested in stimulating T-cells non-specifically, with the endpoint of measuring the production of cytokines and other molecules. Which T-cell stimulant would be most adequate for the task?
If you are plating these in traditional cell culture plates, bind almost any CD3 and CD28 antibody to the plate. Add you PBMCs and incuate at least 48hrsand it will work.
if you are looking for a strong non-specific stimulation you could use Staphylococcal enterotoxin B (SEB) as a positive control (https://www.sigmaaldrich.com/catalog/product/sigma/s4881?lang=en®ion=CA).
