Hello everyone,

I'm planning to evaluate the antibacterial effect of a test substance on bacterial cells by using fluorescent dyes for live/dead staining. Unfortunately, I don't have access to a commercial live/dead staining kit like BacLight. However, I do have Propidium Iodide (PI) and Hoechst 33342 available.

Can anyone kindly share a reliable staining protocol using these two dyes for bacteria (e.g., E. coli, Klebsiella, etc.)?

Any help, papers, or experience-based suggestions would be much appreciated!
