THP-1cells were treated with bacteria , and the three treated group showed two bands on photographic film , while the control group(without bacteria) showed one band . How can I analyze the bands and could you give me some advice?
If Beclin1 siRNA treatment can reduce the densities of both the bands, then you analyse two bands for treated groups and one band for control group using your normal immunoblot analysis software like NIH ImageJ
Is your experiment looking for cleavage of Beclin1 in response to the bacteria, or total Beclin1 expression? It would appear that other papers looking at that protein show a cleavage product: https://www.ncbi.nlm.nih.gov/pmc/articles/PMC3122060/ (Fig 2D).
Some antibodies to Beclin 1 also recognize Beclin 2. Treating cells with bacteria may induce a specific type of autophagy (xenophagy), so try to use siRNA for Becllin 1 and Beclin 2 do distinguish it.
