Hi, I would like to know how will I identify my marine bacteria using conventional method? I have isolated them from marine sediments, and the bacteria was chosen based on how frequent they appear in the MA-plate and by observing the morphological characterisitic of their colonies. I incubated them at 25 degree celcius for 48 hours, and then they were identified using 16s rRNA PCR. However, the plates in the pictures were incubated for 14 days, and I don't know what happened. The 3rd plate with continuous streaking is identified--Mammalicoccus scurii, the two other pictures have the same colony structures in its 48 hours incubation, but different after 14 days of incubation.

I'm having difficulties in finding references to identify them. I have only used marine agar media because other marine selective media are not available in our laboratory. Btw, I'm an undergraduate student conducting my research study and it's my first time to isolate marine bacteria. Thank you so much!
