I have been doing bacterial growth reduction assay of my phages recently. In all the papers, i have seen that O.D of cultures with added phages (at different MOIs) increase at first ( for 1 or 1.5 hours) along with the control (bacterial culture only) and then shows a decrease in O.D.
Wheras in my case, no increase in O.D is obtained in cultures having phages at different moi.
Secondly, there is no difference in O.D between different MOIs (0.01, 0.1, 1, 10). There is a flat line along with the x axis having values as low as 0.002, 0.004, 0.005 etc. when for control is at 0.9, 1.2, 1.3
I don't know whether my results are right or not.
If you are using too high of an MOI, you might be killing off all the host bacteria after only one round and not really see growth followed by lysis. However if you are truly using an MOI of 0.01 that should not be an issue (are you sure about your numbers) and maybe there is something toxic in your lysate.
1.Check your bacterial culture again.
2．Dilute your lysate and try again.
