If I treat dH20 with 0.1% (v/v) DEPC and autoclave it, is the water still RNase free and can I use it as RNase-free water? Or is it ONLY ddH20 with DEPC and NOT autoclaved that can be used as RNase-free water?
Autoclaving dH2O treated with DEPC helps to ensure that all of the DEPC has been inactivated and that the water is truly RNase-free. The heat from the autoclave will also inactivate any RNases that may be present in the water.
