Hey guys,
I want to measure IL-8 secretion from A549 cells when coincubated with Aspergillus fumigatus. I'm using an ELISA for this. At which timepoint is the highest secretion of IL-8 so I might see the biggest difference between different fungal mutants?
I would appreciate any help from you.

In fact A549 cells almost do not secrete the IL-8 without any stimulation. When I was measuring the IL-8 in the medium via ELISA I have changed the medium after treatment, left the fresh one for 24h (to give cells the time to secrete), then collected the medium and run ELISA.
