I am seeking your assistance regarding the growth of Candida albicans ATCC MYA-1023 in 96-well microplates using RPMI 1640 medium (powder, Thermo Fisher, Cat. No. 31800022) supplemented with MOPS (final concentration 0.165 M). The initial inoculum was 1 × 10⁷ CFU/mL.

After 48 h of incubation at 35 °C (200 µL per well, static, flat-bottom 96-well plates), the RPMI 1/1000 dilution showed the highest OD₅₃₀/₆₀₀, approximately 0.21.

We were expecting OD₅₃₀ values in the range of ~1–1.5 after 48 h, but the readings only reached 0.21. Could you please advise on potential reasons for this discrepancy or suggest troubleshooting steps?
