I am working with a mitotic protein and its interaction with mitotic kinases. We use nocodazole to catch the cells at different stages of cell cycle (prophase, metaphase, anaphase and telophase)
For the detection of mitosis we use histone H3 but we need proteins specific to each phases of M seperately, prophase, metaphase, anaphase and telophase.
one option would be to look at the localization of CPC components (see paper below). This complex dramatically re-localizis during mitosis alowing you to indentify the distinct phases.
