My understanding of the subject indicates (experts may correct me if I am wrong), that the desired mouse model described by you may not be of much help in future research. If the mouse has a functional immune system along with the human immunoglobulin expressed in its body, then it will be hard to differentiate whether the effect is due to human or mice immunoglobulin.

I would rather recommend to use a immunocompromised mouse if you wanna study the effect of human immunoglobulin  in a particular system.

Thank you for your comment. The presence of human IgGs in such mice serves only a purpose of inducing tolerance towards human or humanized mAbs whose immunogenicity you would like to test. In fact this is to avoid xeno-type response towards human immunoglobulins.

Out of curiosity, I have a question:

If mouse immune system is functional too, then how will someone differentiate if the effect is due to humanized mAbs or due to some mouse IgG having specificity similar to the humanized mAbs? Please clarify.

You want to assess the immunogenicity of specific therapeutic humanized antibodies? As far as I know, this is the only available mouse model for such a purpose. Maybe you could screen your antibodies with in silico models ?

thank you for your reply. Apparently there is another one available that is called OmniMouse. Unfortunately that one is also patented and rather used for generating human mAbs and not as a immunotolerant model for testing biotherapeutics. Perhaps I can try neonatal immunization with proteins to obtain a model. All the best,
