I am trying to culture NK92MI cells, but they seem to be extremely small. I tried to subculture them in T25 flask (as described by ATCC ~200 000 cell/ml, in the complete growth medium they recommend), and first time they seemed to proliferate, according to cell counting numbers, but they did not formed rosette-like aggregates as they should according to ATCC, and after a while the proliferation stopped but they did not die. Since then I use 24-well-plates, and I could find some aggregates, but they are not frequent, and the cells which are not forming aggregates are much smaller than the cells in the aggregates. I do not know if this is normal. Has anyone experienced similar findings? How fast is the proliferation normally?
