I am currently looking for an IgG antibody that has its sequence published and public. I have found that all the antibody sequences that are published are only sequences for the variable heavy and variable light domains, is it standard practice to only publish the variable domains?
Also in the event I want to clone the variable domains into a vector to produce this antibody, do researcher just take the published variable domains and pick whichever constant domain for IgG and add it to the vector?
Yes, usually only the variable domains are sequenced, as these are usually sufficient for antigen recognition. For recombinant production of antibodies, the specific variable domains are combined with the constant domains most suitable for the intended application.
Most therapeutic antibody variable regions are not published. Since it is relatively easy to add a given variable region to a given constant region no one bothers to publish the constant regions, (usually)
The constant region is identical in all antibodies of the same isotype.
