We are preparing a conjugate vaccine. we need to conjugate a 2 protein 21kD and 29kD. we bought a Solution Link brand protein-protein conjugation kit. we found it a bit complicated. we just want to find out if we can use an easier way of getting the measurement of the amino-Oligo Concentration on a Spectrophotometer. we do'nt have a Quartez micro Cuvette and we dont need to use them regularly, hence it doesn't worth buying an expensive item for a single use. we only have the plastic semi-micro cuvette.

For the measurement of oligo concentration, you need to find out sample absorption at 260 nm. Some plastic cuvettes are transparent within this range, others are not. You can check the specification for your cuvettes.
Drop photometers such as Nanodrop or similar from other manufacturers are very convenient for measuring DNA - if you have one, they can all measure 260 nm.
Most oligo manufacturers specify oligo amounts on the label, in this case measurement is not required.

we need to take a readings on 354nm and 348 which we cannot use the Nano Drop to take. we ordered the cuvette now because we had to as there is no other solution. firstly, because the reaction is only about 130uL and we cannot take much to do it in a bigger cuvette and secondly, the wavelength is up to 354 for 4FB linker.
