Monolith-based chromatographic purification using CIM monoliths is perfectly suited for AAV purification. The first step is capture on HIC column in presence of high concentration of salt , for example potassium phosphate (elimination of proteins, small molecules and protein-DNA complexes). Then it is followed by intermediate capture on cation exchanger (additional removal of proteins and DNA). If needed, there is polishing step on anion exchanger, where empty capsids are separated from full ones. The process is scalable from very small volumes (down to 0.1 mL columns) up to 8 L columns.
