Any suggestions on stimulation of sorted memory T cells with peptide-pulsed matute DCs?

I am planning to investigate tumor-specific CD4 T cells upon in vitro expansion(10 days) with the cognate peptide. Is the ratio 1:5 (DC:T cells) sufficient? Apart from IL7 and IL2, should I also use IL15?

I would use either IL-2 or IL-7 but not both. Personally I use IL-7 during the stimulation and during the first 3 days of expansion and then add in Il-15 from then onwards.
