I have followed a protocol to isolate collagen from rat tails to use in cell culture however I now need to quantify the collagen concentration and am having problems with this:
In our lab we usually use Bradford for protein quantification, so I firstly tried this but with no success (no blue colour change even with neat sample) which was highly unlikely as it is a thick liquid that should be very protein rich! I then read around that there may be problems with collagen precipitating in the Bradford reagent, so I tried added a small percentage of SDS (0.015%) as recommended by some comments. Still no success - 0.015% SDS concentration in Bradford turned everything blue, and 0.015% SDS concentration in the actual sample yielded the same results as my first run (no colour change and blue precipitate instead).
I am wondering whether to borrow BCA from another lab to see if I have more success with this - but does anyone have any experience with quantifying collagen specifically and have any recommendations for doing so? I have seen a Sircol red kit that is designed especially for collagen but I only need test a single sample and do not know anyone that uses this kit.
