I already asked a similar question. I am looking for a cell line that can be used as a positive control for Western blot and for activty/inhibition assay for CYP2D6. Primary hepatocytes (PHH) are quite expensive and I am looking for an alternative. HepG2 are not that promising, while literature on HepaRG is conflicting, some papers say it has enzyme levels comparable to PHH, others say it is comparable to HepG2, which is almost null. Any personal experience, literature would be appreciated.

Hi Karim,
Why not express CYP2D6 in a cell line of your choosing? You could stably transfect your cells with a mammalian expression vector. For on/off, you could use a Tet vector. For dose-dependent response, you could use the ecdysone hormone-inducible system.
