Hi Hadar, it depends what you want. Did you use WGA as a tracer, then you have to detect the WGA with a Anti-WGA (I recommand the biotinylated one). If you like to to stain some special tissue structures by WGA then you should use WGA biotinylated. The advantage of biotinylated antibodies or lectines like WGA is, you can detect them with conjugated Streptavidin (fluophores like Alexa 488 or CY3 or enzymes like Peroxidase). a good fixation for lectin staining is Bouin´s fluid followed by paraffin embedding. For staining do a dilution of WGA-biotin (for instance 1:50, 1:100,1 :200, 1:400, 1:800) and you will easily find the best dilution for you specific tissue.
