I am stimulating T cells for a study. When I do my cell count with Tryphan blue ( 10 micro liter of tryphan blue and 10 microliter of cell suspension); do you multiply the cell count by 2? Sorry this is basic but there is an argument about this in the lab.

I assume you are using a hemacytometer to do manual cell count. In this case, yes, you need to multiply the counts by 2 to get actually number.
However, if you are using automatic cell count machines, generally the multiplication is already built into the systems, and you do NOT need to multiply the counts by 2. Better to read the manual of the machines first.
Yes, you do need to multiply the counted cell number by 2, because your cell suspension was diluted by the addition of trypan blue.
