After we shoot our construct using biolistic equipment, the plants grow on the antibiotic medium. However, after 1 month, they become white. We use lettuce flanks for tobacco chloroplast transformation. Any suggestions?

Dear Mostafa
There are two reasons for plant whitening if you confirm the presence of the transgene in plants.
1- The transgene is silenced. I have such experience with Arabidopsis and 35S promoter and after 3 to 4 weeks I couldn't see transgene expression.
2- The concentration of selective antibiotic is very high; for saving your plants,you should decrease antibiotic concentration in selection medium.
