I am experiencing some toxicity troubles during the treatment. Basically, my zebrafish did not survive after 2hs of treatment. I am using 5 microM TAM from a stock dissolved in 100ml conditioned water (1:2000 from stock). I have read several papers using the same concentration of TAM (and the same solvents) for 7-9hs treatment periods, during 3 to 4 days. It is very frustrating to see my zf die.
Stock solution 10 mM TAM in ETOH or in DMSO. Final solvent concentration is 0.05% v/v. I am using Tamoxifen from Carbosynth (CAS Number 10540-29-1).
Please, I would appreciate if anyone have a tip or clue about my problem.
Hola Lucas. Antes de aventurarme a opinar, me gustaría saber de qué edad son los peces. Veo que el metabolismo del Tamoxifen es complejo y en humanos está muy bien descripto. Es el hígado donde se detoxifica y tiene un alto "binding" con proteínas.
