I had tried three times for detection of andrographolide in blood after oral adminstration. Standard in mobile and plasma is detected very well with mobilw phase methanol: water at 223nm with very good linearity i.e 0.99. Can any one help me?

According to A. Panossian: humans Andrographolide reach the maximum plasma levels of approximately 393 ng/ml (approx. 1.12 μM) after 1.5–2 hours. Similarly in this study were used a UV diode-array detection method. I think you should test your limit of detection in yur methods!!

As suggested by Jhon you must check your LOD. But you can also keep into mind following things during development of bioanalytical method-
1. You should use the internal standard which accounts for variability in plasma processing.
2. You may increase the injection volume upto maximum limit if your LOQ is above 400 nm.
