Hi friends!!!

I am stuck with a fish fin explant culture. Each time I am trying an explant from fin tissue, cells will radiate nicely and monolayers can be seen. We found them growing for few days as well. But before it reach confluency cells will start shrinking and dying. After few days all cells will detach from the flask and dye. Even confluent flasks when subcultured using trypsin will not attach to the bottom of the flask and will be in a floating condition (not dead but will be in suspension). We have tried with some attachment factors like gelatin, but didn't help. And this is happening with gill tissue as well. Can anyone suggest anything on this. Why the cells are loosing attachment after few days of growth!!! Will any other attachment factors or growth factors will help!!!
