I have tissue samples that were stored on liquid nitrogen after collection and transferred to a -80 freezer. I tried to extract DNA using the Qiagen blood and tissue kit (which works perfectly for fresh samples and samples fixed in alcohol or CHAOS solution), but the DNA is extremely degraded. I tested the same kit for samples that were not frozen and got good results, so it's not a problem with the kit.

Does anyone have any advice or can suggest a different protocol to obtain high-quality genomic DNA from samples fixed at liquid nitrogen?
