I am working on nutrient optimization for cannabis tissue culture media. I wanted to try adding a tricarboxylic acid cycle intermediate to help with nitrogen assimilation and preventing NH4 toxicity. I have found some literature with related procedures that are lacking in details of how to do this successfully. I tried using alpha ketoglutaric acid at 15mM. Being that it is an acidic buffer, I used ~20mM of KOH and only got to 4.1 pH, which prevented my media from fully gelling. Does anyone have any suggestions? Should I be using a different acid or a salt version of the acid? Is this not a practical media additive and should I just focus on trialing difference nitrogen concentrations? I appreciate your help.
