I realize that the number of white blood cells in any given whole blood sample will vary drastically depending on the health of the donor. However, is it possible to calculate a rough average of how many cells you can recover from a typical buffy coat preparation? We would like to do some chromatin assays that require about 1.1 million cells per sample and want to make sure we can collect enough blood to do everything.
Usually you can obtain ten million PBMC from ten ml of blood drawn from an healthy patient.

Stefania is correct. Assume an average of 1x10^7 per 10ml whole blood when isolating buffy coats. I prefer percoll over ficoll gradient. I have had cleaner isolations with percoll (less RBC carryover). I also recommend cell counting your buffy coats with a hemacytometer and a microscope to confirm your counts. I have experienced the averages of PBMC isolations from 1ml WB can vary greatly between populations and even age. If you count 25% of the samples from your whole subset, you can then more accurately gauge the average PBMC yield for your population.
