Buffer Solution contains Tris-HCl (pH 8.0), KCl, MgCl2, DTT, and EDTA. This problem has occurred during 5 consecutive iterations. The buffer itself is clear and the glycerol itself is clear, but combining them turns them pink. I have EtOH washed all the equipment in advance in case there are dye residues present. Do you think the glycerol is off or another one of the reagents?
This definitely should not be happening. The glycerol must be contaminated. Get some new glycerol and try again.
only thing I can think of is your water.. is it deionised.. Any chance that there a Cobalt salts?
Thank you both for your comments. I did try new glycerol and the result continued to happen. After consulting another biochemist in the department, we determined that it was likely due to metal contamination and the addition of glycerol caused it to chelate, so perhaps Cobalt...? After about 30 minutes on the stir plate, the color disappeared and the buffer became clear.
