the purpose of aGC clmp is to make sure that there is one part of the molecule that does not melt.This part is the GC clamp.then there can be differential melting of the part of the amplimer that may contain changes so that when that part of the mplecule melts the whole amplimer does not fall apart but just stops moving in the gel. Without a gc clamp the smaller low melting amplimer might just completely fall apart and you would never see the difference between the normal molecule and a mutated molecule because everything melted too easily. With the clamp it should be possible to see most changes within the low melting domain melting slightly differently
