Dear all,

I bought a recombinant human MCP-1 protein from Thermo FIscher and trying to optimize the signaling activation by using target cell X. I followed the company's instruction to dissolve the recombinant MCP-1. Briefly, we made 100ug/ml solution by using distilled water. Further dilutions were made by using 0.1% BSA in PBS.

When we treated our target cells with rec. MCP-1 protein then we observed the signaling activation. However, we also found the signal of almost same intensity in our negative solvent control (0.1% BSA in PBS). I am confused that either the activation of signaling is because of MCP-1 or the effect of BSA.

I need your help in order to know any alternative solvent or using heat-denatured BSA? Is heat-inactivation of BSA is the right option?
