In single TCR alpha transgenic mice (the alpha chain was cloned in pTalpha cassette, the mouse strain was obtained on the background on which the original TCR was isolated) we see 2x increase in DN CD3+ thymocytes as compared with wild type mice, but this increase is not due to gamma/delta T cells. The expression of the transgenic alpha chain is fully absent in DN, DP, SP thymocytes and peripheral subpopulations of T cells. Cellularity of thymus in transgenics is decreased (~50% of the thymus of wild type mice). Please, could anybody explain this phenomenon?
