I am working on a in vitro inflammatory model(a co-cultured model) which stimulated with IL-6. After stimulation ,the control and control+IL-6 show no significatnt differences in expersion of proinflammatory cytokines TNF-a and IL-8.
I would like to know your opinion on this result. Iam thinking the model is not working! what do you think?

Are you using an established protocol?
When you say expression, do you mean message or protein?
What are the cells in your co-culture?
At what time point are you taking the samples?
And what is your concntration of IL-6?
IL6 also has anti- inflammatory function depending on the cell context. Do you see a dissociation of your cells from the feeders as a first hint of pro-inflammatory response (if your cells are epithelial). If not, then most likely you won't see a change in the secreted pro-inflammatory cytokines.

I know IL-6 is a ploripotent but when it comes to intestine and liver cells is proinflammatory. I think.
To answer ur question Shen:
- protein,
- After 24 stimulation with IL-6 cells will be lysed.
-intestinal and liver cells
-Different concentrations and all less than 5ng/ml
