Hello all, I am trying to grow anaerobic bacteria (from feces) from a glycerol stock. I was told that the stock needs to stay frozen so it must stay on dry ice. However, I cannot bring the dry ice into our anaerobic chamber because it would mess with the gas mixture. I was told to dig out the bacteria from the stock while at a lab bench. Won't this kill the anaerobic bacteria?
Also, I tried growing my bacteria on BSM agar over the weekend. I got 2 of them to grow out of 30+. Will it just take longer? I'm not sure if I took enough from the glycerol stock. Does anyone have a better technique for doing this?

If you don't have a chamber, you could try this: https://www.bd.com/en-us/offerings/capabilities/microbiology-solutions/environmental-systems/bd-gaspak-systems-and-accessories. However, I think if you are trying to isolate very strict anaerobes, I'm not sure this would work well.
I finally ended up with an answer to this question. I took a cold plate/block and kept it in the -80C freezer. Then I bring it into the chamber with me and just do what I need to as fast as I can. The plate will warm up rather quickly, so you can keep it on dry ice until you are ready to bring it into the chamber.
