I have a question about the count of 3D spheroids.
How can I count the number of spheroids starting from the number of seeded cells? After 3 days colture I count spheroids that have 100 um of diameter. Anyone suggest me a simply matematical method to count a number of spheroid (MTCs) formation?

Could you add more details:
- What kind of images do you get? Modality, 2D, 3D, etc? (this will impact the detection algorithm choice)
- Could you post examples?
- Do you need a precise count or could you just use a sampling approach? Would your spheroids be (relatively) uniformly distributed in your ROI?

I have 3D structure in Alginate scaffold. and I seeded 1x10^5 cells in a 24 well plate. I not have a precise counter but i use a samplig approach.
An automated counting option may be helpful to you here. The GelCount for example can unbiasly count only spheroids you want (ie those over 100um) and not count the rest. This would provide a precise measurement. Cheers.
