Will the peptides, used for PBMC activation, bind directly to the cell surface MHC II molecule or are they internalized, processed and then presented by MHCII?
A good reference would be helpful. Thank you.

Depends on the peptide sequence, depends on the APC doing the presentation. 15mers canonically would require some processing step before they can bind to the MHC, but the attached paper would indicate that 18-20 aa is the optimal length of peptide bound to MHC.

Thanks Blake for the response and the article.
I found an article as well. The paper describes the peptide trafficking to cytosol and endosomes. Wonder how the peptide enters the cells though.
