15mer peptide fragments are used for activation of PBMC in Elispot. Are these peptides internalized?

 Will the peptides, used for PBMC activation, bind directly to the cell surface MHC II molecule or are they internalized, processed and then presented by MHCII? A good reference would be helpful....
