I am using a transgenic fish that fluoresces in the liver. When injected with a particular morpholino I can knockdown the fluorescent signal, however, the embryo's development is often severely affected. Can anyone suggest a specific zebrafish liver marker that I could get an antibody for, to prove the liver is still present i.e. that the morpholino injection is causing knockdown of a specific gene, not disrupting development to the point where the liver is effectively not present?
The prox1 antibody will label the liver. However the antibody search at ZFIN (http://zfin.org/action/antibody/antibody-search) will give details of antibodies reported to label the liver (including Prox1).
Eventually you can use ISH instead of IF/IHC with specific liver markers (such as Fabp10a, like probably the transgenic embryos you are injecting)
Ok, I guess I'm old school. The liver is a big organ - even in larvae. I'd just section the fish and do an H&E stain.
Instead of an antibody, you could also breed your transgene into the 'absolute' line, which is an adult viable transparent double mutant of mitfa;bdnrb1a. You can see the liver in live fish and won't have to use a marker. I concur with Robert's response regarding ZFIN antibody search. There are probably several well characterized markers out by now. One marker I know about are zn-5 monoclonal antibodies which label mainly CNS tissue, but also pancreas and liver.
You can do RNA insitu hybridization for LFABP (Liver Fatty Acid Binding Protein) gene at 3 dpf zebrafish embryos to prove that liver is still present in those injected embryos. LFABP is very specific to liver. (only liver)
