I would like to investigate the role of certain amino acids for survival of S. cerevisiae cells. To this end, I would like to deplete the cells for an amino acid of interest by incubating in media w/o this amino acid.
I have already searched the literature, but most papers describe general nitrogen starvation by incubating in SD-N for about 3 h, which reduces the total cellular amino acid content to about 25% of the non-starved control (e.g. Onodera & Ohsumi, JBC, 2005).
Could I deplete specifically e.g. the intracellular histidine pool if I incubated a strain which is auxotroph for histidine in His-free media? How long should I incubate the cells?
Any advice on protocols or literature regarding this topic would be greatly appreciated.
