Several studies have shown that PPP decreases the signal of light transmission aggregometry and this effect is not related, either by the volume expansion or by ADP-released during the centrifugation step.
Questions:
1) Do you know the mechanism that PPP decreases platelet aggregation?
2) Is there any study showing similar effects on platelet activation?
3) Would not be more physiological to use PPP instead of Tyrone buffer and assume that platelets are less responsive to activation?

I have not found that PPP decreases platelet aggregation. If, however that PPP was isolated from blood anticoagulated with ACD, there may be an effect. You can dilute with Tyrode's buffer to reach the required platelet count. But, you also must dilute the PPP negative control to the same extent to obtain an accurate blank (100% aggregation) reading in the aggregometer.
