I have got the plasmid that is not in the bacterial stub but dried droplet form. We do not have any experience in dealing with this kind of plasmid. Is it possible to make a solution from this form?
Where did you get the plasmid from? The protocols for making a solution from the droplet should be obtained from the organization who constituted it as such. Is it plasmid DNA or a strain? Often both are sent on filter disks. If it is a bacterial strain (carrying a plasmid) then use sterile forceps and just streak across an appropriate selection plate. Or you can just drop into a tube with a few ml of appropriate growth media. Let grow and be sure to pick single colonies for further propogation and storage.
If it is pure plasmid DNA then put the filter into a small microfuge tube, add some TE or similar (enough to saturate the filter and leave some liquid around - maybe 50-100ul). Then remove a microliter or two and use for transformation.
