My current knowledge is that with certain growth factor or stimulants (pokeweed mitogen), lymphocyte population could be expanded in cell culture. Adding IL3 could help basophil's survival in cell culture. But are they (basophil, eosinophil, monocyte) able to proliferate in cell culture? If so, what is the condition? Our lab is developing an algorithm to recognize WBC subsets, so we need large amount of purified WBC subsets to facilitate the machine learning process. Any information would be appreciated.

As you mentioned, IL-3 acts as a basophil growth factor. However, even in IL-3, basophils do not survive beyond 2 weeks. IL-3, unlike several other cytokines enhance basophil survival by blocking caspase-3 cleavage of bcl-2. You may refer to the articles attached below for more information.

The investigators claim that they and other groups have shown that interleukin-5 (IL-5) maintained the viability of mature eosinophils in an in vitro liquid culture system. Mature eosinophils did not proliferate but their survival was maintained in the presence of IL-5. In the absence of human IL-5 (hlL-5) mature eosinophils succumbed after 4 days, while in the presence of hlL-5 they survived up to 10 days.

Regarding monocyte, you may refer to the article attached below. The investigators claim that blood monocytes from healthy volunteers were isolated by Ficoll-Isopaque centrifugation and cultured (together with lymphocytes) in medium 199 with 20%, heat-inactivated newborn calf serum in a Teflon culture bag. Quantifiable data on survival showed that up to 21 days of culture, approximately 40% of the initial number of monocytes were still viable. Such cultures could be maintained for more than 8 weeks without refeeding.
