I'm using methylcellulose as an overlay for PA (before I always used Agarose) does anyone have suggestions of
#1 how to get this stuff dissolved
#2 be able to pipette so you can make a 1:1 solution with MEM
I tried today to pipette my 2% solution of methylcellulose and it was so viscous that it was almost impossible to pipette and be accurate.

What I usually do is to autoclaved the powder (exactly and then under the hood add to a DMEM or MEM. And put in the 4°C with a shaker for 1 night or 1 day, even the weekend and then you have your methylcellulose ready to add FS and antibiotic and ready tu use.

I usually do the same as Costanza. It works fine.
Indeed it is very difficult to be accurate during pipetting when working with methylcellulose. If it is suitable for you, I recommend to try with carboxy methylcellulose at higher %.
