glucose (50%) was injected IP, then the glucose in blood was measured.
The problem was, the glucose in bood of rats did not increase very fast and high.
For example, the glucose in some rats was more than 600 mg/dl, but in most other rats it was only 200-300 mg/dl 15min after being injected gluocose. which value should be normal or right?
in addition, some people argued that the glucose should be the highest in 15 min but our data showed the highest in 30 min.
I am not sure whether we injected in the wrong position.the needle we used was 23 G.
would you give me some advice on this? I will apreaciate that.

The dose was 2g/KG, the weitht of rats were around 200-300g,which means the injection amont was 0.8-1.2 ml 50% gluocose per rat.
In mice, I get around 400 to 500 mg/dl after 20% glucose injection at 1 g/kg body weight at 15 to 35 min post-injection. I also found that way you handle the animals affects a lot How were the baseline values? Another thing, its not golden rule that glucose should be highest at 15 min. It can be between 15- 35 min but it would be surprising if does not increase after glucose injection.

For the baseline values, do you mean after fast overnight and before gluocose injecting? at that point, the glucose was around 70-90md/dl.
But I am not sure how you think of my data. Do you mean in 30 min, 200-300 md/dl is not high enough?

The glucose dose at 2gm/kg is on the low side from our experience but it all depends on your target peak level of blood glucose - in other words, the reason for your doing the experiment.
Some inter-animal variation in blood glucose levels in a group of normal animals is expected, which can also shift the peak level in the range of 10-30min. However, a difference between >600mg/dl in some versus 200-300mg/dl in others is unexpected. It is possible that while injecting glucose into the peritonium, you ended up injecting glucose in one of the internal organs, which would delay its appearance in blood and lower the peak level. In some cases, for example, injecting into the bladder, you will see very little change.
If you wish, I can post one of our graphs from normal Wistar rats... which will show what I have stated above.

We tend to challenge our experimental animals rather seriously and use a dose of glucose ranging between 3.0 - 4.0 gm/kg. You may have someone restrain rats for you so there is no movement where glucose ends up in an internal organ. Size of the needle is only as important as your comfort level with it, the process needs to be quick - I tend to use 18G for larger rats (300gm+) and 20 or 22G for smaller rats. You have to ensure that glucose is delivered in the peritoneal cavity and not into the intestines, bladder or any other site.
Rather than including unpublished data, I am including something that was published a few years ago:
The glucose in normal rats can be highest between 15-30 min, but the glucose in some rats more than 600 mg/dl versus 200-300 mg/dl in other rats is unexpected 15 min after injection of glucose. You may not perform a precise intraperitoneal injection (injection of glucose into the peritonium).

I saw your data and also adjust the dose to 4 g/kg bw. Most of the data looked normal then, but another problem happened: the biggest value of our glucose meter is only 600 mg/dl. could you tell me the lot number of your meter which looked have a larger range of measurement.
